人肝星形细胞说明:


       肝脏星形细胞是小叶内结缔组织细胞,呈现肌成纤维细胞样或脂肪细胞形态。他们参与肝脏细胞外基质的稳态、修复、再生、纤维化以控制视黄醇的代谢、存储和释放。肝脏受伤后,肝脏星形细胞转化成肌成纤维细胞,并且是纤维化的肝脏中I 型胶原的主要来源。除了以上特征,已发现在疾病状态和肝内门静脉高压的发病过程中,肝星状细胞通过细胞收缩调节肝脏微循环、星形细胞的增殖、迁移和趋化因子的表达参与了肝脏炎症和纤维化的发病过程。星形细胞支配电压激活的钙离子流,表达低亲和力的神经生长因子受体p75,在神经生长因子的刺激下发生细胞凋亡。因此,星形细胞激活的分子调控在将来有助于治疗纤维化肝,并且能减少慢性肝病患者的发病率和死亡率。   

        人肝脏星形细胞(HHSteC)提取于人肝脏组织,原代冻存。每管含有细胞数>5×105cells/ml,此细胞通过对Desmin, alpha-Actin和Fibronectin免疫荧光染色验证,经测试不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌。细胞可以达到15倍增。

Hi, We are interested in purchasing human hepatic stellate cells (#5300). I have few questions. are these cells isolated from a single donor? How much quiescent phenotype these cells do maintain? Do you recommend any specific culture dishes to grow these cells as well as to study quiescent HSCs?

Hi Pradip,
Hepatic Stellate Cell quiescence and activation is a complicated matter. In general, cells expressing high levels of alpha-smooth muscle actin should be activated and cell not expressing alpha smooth muscle actin should be quiescent. Stellate cells are also able to switch from the activated state back to the quiescent state under certain circumstances. Stellate cells are known as reverted stellate cells at this time and this is an intermediate phenotype.  
I would recommend using the cells as quickly as possible for experiments, however the cells will be proliferative. If you need a completely quiescent state I would recommend reading the following paper as it indicates how to revert hepatic stellate cells back to the quiescent state:

Is there a preferred protocol for siRNA transfection of these cells?

We do not have a preferred protocol for siRNA transfection of Human Hepatic Stellate Cells. We do offer a variety of transfection kits for a variety of cell types (0903, 0933, 0913, 0943, 0963, 0953, 0923, 0973). The most useful one may be the Smcfectagen (0953) but it has not been tested with Human Hepatic Stellate Cells.

We have tested your HSC cells and our collaborators have ordered a larger batch of them. I can say that the quality was quite high. At the moment we need another lot of hepatic stellates from new adult donor with no major pathologies. Could you please let me know if you have anything in stock at the moment? Also, if you ever get stellates from a donor with liver pathology (NASH, non-viral hepatitis, cirrhosis etc.) we would like to test them as well. Could you please keep us informed? Thank you.
