人角膜上皮细胞说明
人角膜是唯一的无血管的组织,具有透明的特性和合成许多蛋白的功能。角膜包含三个不同的细胞层,外层是上皮细胞,内层是内皮细胞,中间层基质层(角膜细胞)。角膜上皮细胞能参与先天性免疫,它能感应病原体存在并发出信号从而激活角膜防御系统。还能高效表达醛脱氢酶,防止UV-和4-羟基壬烯醛对细胞造成伤害。角膜内皮细胞增殖是由细胞因子控制的,比如说表皮生长因子,它能激活上皮细胞深层的同源受体。体外培养角膜内皮细胞已经可以替代检测动物眼睛的产物而用于毒理学检测和人角膜上皮细胞分化的分子机制的研究。
人角膜上皮细胞(HCEpiC)提取于人角膜组织,于原代冻存。每管含有细胞数>5×105 cells/ml,HCECIC的特点是卵石形态无血清培养和免疫荧光与特异性抗体CK-18和-19。HCEIC对HIV-1、HBV、HCV、支原体、细菌、酵母菌和真菌均为阴性。
