MET-5A是一种人间皮细胞系,源自正常成人胸膜液中的间皮细胞。这些细胞在转染过程中使用了含有SV40病毒早期区基因和RSV长末端重复序列(LTR)的pRSV-T质粒,采用DNA锶磷酸盐共沉淀法。转染后的细胞经过连续传代,并每周更换培养基,经过28天后,对转化的克隆进行了独立培养。pRSV-T质粒的关键作用是通过SV40大T抗原抑制细胞周期调控蛋白,如p53和pRb,使MET-5A细胞能够逃过正常的细胞周期检查点,保持持续的增殖能力,从而实现了细胞的永生化。
MET-5A细胞表现出间皮细胞的特征,包括对波形蛋白(vimentin)、细胞角蛋白(cytokeratin)和SV40T抗原的阳性染色表达。这些细胞同时表达间皮细胞和上皮细胞的某些标记物,反映了它们的间皮细胞起源和上皮样形态。 |
