产品介绍

SW480是一种人结肠腺癌细胞系,SW480和SW620均源自同一位男性患者,其中SW480是原发性结肠腺癌。最初是从一位50岁白人男性患者的直肠腺癌组织中分离得到的。这个细胞系在癌症研究中非常有用,尤其是在研究结直肠癌的生物学行为和药物反应方面。该细胞的CSAp和结肠抗原3阴性,角蛋白免疫过氧化物酶染色阳性。该细胞的p53基因第273位密码子的G->A突变导致Arg->His替代,第309位密码子的C->T突变导致Pro->Ser替代。该细胞p53蛋白表达水平升高,癌基因c-myc、K-ras、H-ras、N-ras、myb、sis和fos的表达呈阳性,但未检测到癌基因N-myc的表达。此外,该细胞不表达细胞基质裂解酶Matrilysin(一种与肿瘤入侵相关的金属蛋白酶)。有报道称该细胞表达GM-CSF。此外,该细胞K-RAS蛋白第12位氨基酸Gly突变为Val(c.35G>T),常被用于PCR检测该位点突变的阳性对照。该细胞是研究结直肠癌的重要工具,为科学家们提供了一个模型系统,用于研究疾病的进展和开发新的治疗方法。

注意事项:
1.该细胞使用的L-15基础培养液由磷酸盐和游离碱氨基酸代替了碳酸氢钠缓冲体系。该培养基必须在没有二氧化碳平衡的环境中培养细胞,不能在普通的5%二氧化碳细胞培养箱中培养,否则培养液颜色会明显变黄。
