产品介绍

人乳腺细胞MCF 10A是一种非致瘤性上皮细胞系。上皮唾液酸粘蛋白(epithelial sialomucins),细胞角蛋白(cytokeratins)和乳脂肪球抗原(milk fat globule antigen)阳性。MCF 10A在胶原蛋白中呈现三维生长,并在融合时形成圆顶状形态。迄今为止,细胞未显示终末分化或衰老的迹象。该细胞系对胰岛素、糖皮质激素、霍乱肠毒素和表皮生长因子(EGF)有响应。通过电子显微镜观察,该细胞呈现管腔导管细胞的特征,但不呈现肌上皮细胞的特征。培养液中的钙含量对细胞的形态影响很大。

注意事项:
(1)该细胞较难消化,注意延长消化时间,消化到细胞收缩变圆,轻拍培养瓶侧边,细胞可以滑落方可终止消化。
(2)该细胞贴壁较慢,传代后贴壁24~48小时后再进行后续操作。
(2)建议使用Corning的cellbind细胞培养瓶(货号:3289)进行细胞培养。
(3)使用0.25%胰酶消化后,加入几倍体积的完全培养液稀释后,1000rpm/5min,去除胰酶后加入完全培养液进行传代。
